The Royal Canadian Mounted Police say the attacks took place at several locations, including the James Smith Cree Nation and the village of Weldon, northeast of Saskatoon.
Initial evidence suggests that the victims may have been randomly attacked, but the police did not specify the number of injured or whether anyone had died.
For his part, Diane Shir, a resident of Weldon, said she was in her garden on Sunday morning when she noticed emergency teams two buildings away.
Explaining that her neighbor, a man who lived with his grandson, was killed.
She did not want to disclose the victim out of respect for his family.
Adding, "I am very upset because I lost a good neighbor."
She said that businesses in the village of about 200 people are usually closed on Sundays, but stores in larger neighboring communities were closed as a precautionary measure.
The suspects were being sought when fans in Regina were arriving for the sold-out annual Labour Day game between the Saskatchewan Roughriders in the Canadian Football League and the Winnipeg Blue Bombers.
Regina Police said in a press release that, with the help of the Monties, they were investigating on several fronts to locate and arrest the suspects and "deployed additional public safety resources across the city, including the football game at Mosaic Stadium."
Explaining that the alert initially issued by the Melfort RCMP around 7 a.m. was extended after hours to cover Manitoba and Alberta, where the suspects remained at large.
The suspects were described as Damian Sanderson, five feet seven inches tall and 155 pounds, and Miles Sanderson, six feet tall and 200 pounds.
Both have black hair and brown eyes and may be driving a black Nissan Rogue with license plate 119 MPI.
The Saskatchewan Health Authority said many patients are receiving treatment at several locations.
Spokeswoman Ann Linman said in an email, "Additional staff have been called in to handle more victims."
Mark Audan, spokesperson for air ambulance, said two helicopters were sent from Saskatoon and another from Regina.
Audan said two transported patients from the scene to the Royal University Hospital in Saskatoon, while the third transported a patient to the Royal from a hospital in Melfort, a short distance southeast of Weldon.
Audan said that due to privacy laws, he could not disclose information about the ages, genders, or conditions of the victims.
He said other patients were transported by ground ambulance to hospitals in Prince Albert, Nipawin, and Melfort, but their number is unknown.
Adding that the second helicopter sent from Saskatoon carried a transport doctor to the scene to assist with operations on the ground, but he does not know the location.
